These perks are good choices after you secure your first upgrades. They might not be the absolute most important choices for every single run, but they give you solid utility. They can make your life much easier depending on how you like to play in 33 Immortals.

Best A-Tier Perks
The Light Bringer perk lets you start your runs with extra bones in your pockets. You use bones to buy Keys, heal up, or grab Teleport Stones. You can always find bones in chests anyway, but a free head start at the beginning of a round is a nice bonus.
