You can't use Custom Runs in Slay the Spire 2 right away. You can unlock the mode in the early-access version by beating Act 3 three times in normal play. The most important thing is that, so the quickest way is not to look for rare unlock tricks.
It just means finishing Act 3 three times as quickly as possible. The current guide coverage also says that Custom Runs is a separate mode for modifiers, experiments, and setting up challenges after you have won.

The Quickest Way to Get the Three Clears
