To reach Hamamatsu Castle, you need to follow the main path laid out before you. The trail acts as a guide, leading you directly toward a cinematic sequence that introduces the area in Nioh 3.
As you move forward, you will notice a massive, dark creature in the sky that looks like the one from Edo. To get closer to the castle, loop around to the left side of the area

Preparing for the Climb
Before you get too far into the mission, stop at the Shrine to save your progress. Use your points to Level Up and work on making your Ki Recovery Speed faster so you're ready for the fights that are coming.
As you advance, you will encounter a very tough enemy. It is best to use a Sneak Attack to deal a large amount of damage early.
Keep your health above fifty percent during this fight, as one full combo from this foe can take you out instantly. If you fall, remember to pick up your Grave before heading back to the Shrine.

Navigating the Castle Grounds
The air feels heavier as you get closer to Hamamatsu Castle. The path forward can feel like a puzzle, but you can find your way by following the enemies in Nioh 3.
If you see guards, you are likely heading in the right direction. Use your Bow to clear out Archers from a distance so they do not surprise you. When you see another Shrine up ahead, you will know you are on the correct track.

Breaking the Barrier
Near the castle, you will face a trial where enemies attack in waves. Be careful, as these enemies can lower your maximum health when they hit you, making it impossible to heal that portion of your bar for a while.
This is a good time to use your special ability, like the Cat spirit, to stagger your foes. For the final enemy in the wave, activate your special mode to finish them quickly in Nioh 3.
When the area is clear, you can break through the wall and run past any guards that are still there to get to the final cutscene at the castle gates.
